Addition of fungal inoculum increases seed germination and protocorm formation in a terrestrial orchid

چکیده

Because orchid mycorrhizal fungi are important for seed germination, addition of germination-supporting fungal inoculum to the soil can be an efficient way improve in situ germination terrestrial orchids, but empirical evidence is still largely lacking. In this study, we used inoculation experiments test hypothesis that increases Gymnadenia conopsea under natural field conditions. China, species has suffered dramatic declines abundance due habitat loss, grazing and over-collection, while observational studies have shown very low recruitment seedlings into populations. Previous research strain GS2 (Ceratobasidiaceae) was able support vitro Seed packages were bury seeds with without assess germination. total, 11.96 ± 2.65 % had testa ruptured when added packages, 10.63 3.32 germinated protocorm formation stage, 5.24 2.71 grew differentiation 3.16 1.63 finally formed early seedlings. contrast, no observed control groups inoculum. We isolated associating germinating confirmed supported by GS2. addition, several other from seeds, including four Ceratobasidiaceae strains known vitro. Our results highlight potential induce orchids conditions may useful developing more symbiotic protocols restoration conservation especially situations where severely limited.

In vitro asymbiotic germination of immature seed and formation of protocorm by Cephalanthera falcata (Orchidaceae).

BACKGROUND AND AIMS Many Orchidaceous species are threatened globally by development and over-collection from their natural habitats for horticultural purposes. Artificial propagation from seeds is difficult in most terrestrial orchids native to temperate regions. Seed production is another limiting factor in the artificial propagation for these species because of the lessened probability of po...

Effects of culture medium and supplementation on seed germination, protocorm formation and regeneration of some Phalaenopsis hybrids

An efficient protocol is suggested for in vitro culture of five Phalaenopsis hybrids obtained by hand-cross-pollination of three commercial hybrids Calgary, Ankara, and Kendall. Four nutrient media- namely half-strength Murashige and Skoog, Knudson, Phytamax containing activated charcoal, and Mitra– once supplemented (with coconut water, peptone, or both), and once without any supplement were c...

Fungal specificity bottlenecks during orchid germination and development.

Fungus-subsidized growth through the seedling stage is the most critical feature of the life history for the thousands of mycorrhizal plant species that propagate by means of 'dust seeds.' We investigated the extent of specificity towards fungi shown by orchids in the genera Cephalanthera and Epipactis at three stages of their life cycle: (i) initiation of germination, (ii) during seedling deve...
